Description:
This recombinant human adenovirus type 5 vector expresses a codon-optimized
Streptococcus pyogenes Cas9 nuclease carrying the D10A mutation (nickase
variant), with co-expression of DasherGFP via a T2A self-cleaving peptide under
the control of the CMV promoter. The vector is designed for use in combination
with guide RNAs (gRNAs) to enable efficient and precise genome editing in
mammalian cells.
The Type II prokaryotic CRISPR/Cas system is a powerful platform for targeted
genome engineering. Cas9 (CRISPR-associated protein 9) is an RNA-guided DNA
endonuclease derived from the adaptive immune system of Streptococcus pyogenes.
In CRISPR/Cas systems, small RNAs (guide RNAs, gRNAs) direct Cas9 to
complementary DNA sequences, where it introduces sequence-specific strand
breaks.
Because guide RNAs can be easily designed to target virtually any genomic locus,
the CRISPR/Cas9 system provides a faster, simpler, and more robust alternative
to traditional genome-editing technologies such as TALENs and zinc finger
nucleases, and is widely used for gene knockout, knock-in, and functional
genomics applications.
