Description:
These recombinant human adenovirus type 5 (E1/E3 deleted) vectors express a
codon-optimized SpCas9 nuclease, with select constructs driven by the CMV
promoter. They are designed for use in combination with guide RNAs (gRNAs) to
enable efficient genome editing in mammalian cells.
The Type II prokaryotic CRISPR/Cas system is a powerful platform for targeted
genome engineering. Cas9 (CRISPR-associated protein 9) is an RNA-guided DNA
endonuclease derived from the adaptive immune system of Streptococcus pyogenes.
Guided by a programmable single-guide RNA (sgRNA), Cas9 introduces
sequence-specific double-strand breaks at defined genomic loci.
Because guide RNAs can be readily designed to target virtually any DNA sequence,
the CRISPR/Cas9 system provides a faster, simpler, and more robust alternative
to traditional genome-editing technologies such as TALENs and zinc finger
nucleases, making it well suited for gene knockout, knock-in, and functional
genomics applications.
