Description:
AAV9-CAG-DIO-mCherry is a pre-made recombinant AAV vector of serotype 9 that
enables Cre-dependent expression of mCherry under the control of the CAG
promoter (also known as the CBA promoter), as illustrated in the schematic
below.
mCherry is a bright, monomeric red fluorescent protein that matures rapidly,
allowing visualization of expression shortly after transcription is activated.
It exhibits strong photostability and resistance to photobleaching, making it
well suited for both in vitro and in vivo applications.
In the DIO (double-floxed inverse ORF) configuration, the mCherry transgene is
inserted in the reverse orientation relative to the 5′ promoter and is flanked
by oppositely oriented loxP and lox2272 sites. In the absence of Cre
recombinase, the transgene is not expressed. Upon Cre expression, permanent
Cre-mediated recombination inverts the transgene into the correct orientation
(“flip-exchange” or FLEX), resulting in robust mCherry expression.
This system is also commonly referred to as DIO, Cre-ON, FLEX-rev, FLEX-ON/FlexON,
or AAV-DIO.
