Description:
AAV2-CAG-mCherry-GFP-LC3 is a pre-made recombinant AAV (rAAV) vector packaged
in serotype 2, containing AAV2 capsid proteins and AAV2 inverted terminal
repeats (2× ITRs). This vector expresses the mCherry–GFP–LC3 fusion protein
under the control of the CAG (also known as CBA) promoter and serves as a
specialized tool for studying autophagy.
AAV2 exhibits relatively broad tropism when administered in mice or used in
vitro. Early autophagic vesicles have a near-neutral pH (approximately 7.4),
under which both GFP and mCherry fluoresce, producing a merged yellow signal.
Upon fusion of an autophagosome with a lysosome—a highly acidic organelle with
a pH of approximately 4.5–5.0—an autolysosome is formed. Because GFP is
acid-labile, its fluorescence is rapidly quenched in acidic conditions,
whereas mCherry is acid-stable and remains fluorescent. Consequently, red fluorescent puncta
indicate late-stage autophagic vesicles (autolysosomes), while yellow signals
represent early-stage autophagic structures. This product is supplied in a
