AAV8-CAG-DIO-ChIEF-tdTOMATO
is a pre-made rAAV in
serotype 8 which will Cre dependently over-express DIO-ChIEF-tdTOMATO
driven under CAG promoter (also known as CBA promoter) as illustrated as the
cartoon below.
ChIEF is a hybrid of channelrhodopsin 1 and 2 with additional L170I mutation
(ChR1 numbering) or at L131I (ChR2-numbering), leading to large currents in
oocytes and HEK-cells and almost wild type like open state life time.
ChIEF function as light-gated ion channels and very useful for many
bioengineering and neuroscience applications such as photostimulation of neurons
for probing of neural circuits. Using tdTOMATO tagged ChIEF, light-stimulated
axons and synapses can be identified in intact brain tissue. This is useful to
study the molecular events during the induction of synaptic plasticity. ChIEF
has also been used to map long-range connections from one side of the brain to
the other, and to map the spatial location of specific inputs on the dendritic
tree of individual neurons.
In the DIO scenario, the transgene of interest is inserted in reverse
orientation relative to the 5' promoter and is flanked by oppositely oriented
loxP and lox2272 sites. In the absence of Cre expression, the transgene
will not be produced. In the presence of Cre expression, the transgene will be
"Flip-exchanged" or FLEXed, leading to the expression of the transgene. This is
due to a permanent Cre-mediated recombination/inversion of the flanked transgene.
This arrangement is called DIO (double-floxed inverse ORF), Cre-ON, Flex-rev
(reverse), Flex-ON/FlexON, or DIO-AAV/AAV-DIO (double-floxed inverse ORF in AAV).
