Description:
GCaMP is a genetically encoded calcium indicator (GECI) engineered from a
fusion of green fluorescent protein (GFP), calmodulin (CaM), and the M13
peptide derived from myosin light-chain kinase. Upon binding intracellular
Ca²⁺, GCaMP undergoes a calcium-dependent conformational change that increases
the proximity and fluorescence efficiency of the GFP moiety, resulting in a
robust increase in green fluorescence. This fluorescence change enables
real-time monitoring of neuronal activity, action potentials, and
receptor-mediated calcium influx in living cells and organisms. A key
advantage of GECIs is that they can be genetically targeted to specific cell
types, allowing stable, cell-type-specific calcium imaging in vivo.
GCaMP6 represents a next-generation, ultra-sensitive class of GCaMP indicators
and significantly outperforms earlier variants in both signal amplitude and
detection accuracy of cytosolic Ca²⁺ dynamics. Targeted mutations within the
calmodulin domain of GCaMP6 have produced variants with distinct calcium
binding and fluorescence decay kinetics, including fast (GCaMP6s), medium
(GCaMP6m), and slow (GCaMP6s) response profiles, enabling researchers to
select the optimal sensor for their experimental time scale.
AAV9-CaMKII-GCaMP6s is a pre-packaged recombinant AAV vector of serotype 1,
containing an AAV9 capsid and AAV2 inverted terminal repeats (ITRs). It drives
robust expression of the fast-kinetics calcium indicator GCaMP6s under the
control of the CaMKIIα promoter, ensuring neuron-specific expression,
particularly in excitatory neurons. This vector is supplied in a ready-to-use
format for in vivo and in vitro neuronal calcium imaging applications.
