LipoD293™ In Vitro DNA Transfection Reagent

We transfected 293T derivatives that have an extra Large T transgene (293TT) used for papillomavirus pseudovirion production. The initial transfection was performed with the entire sample you sent, i.e., 60 ul of LipoD293 15 ug of GFP carrying plasmid, and 20 ug of capsid plasmids (total of 35 ug of DNA). We observed greater than 60-70% transfection on a 70% confluent T175 flask. This level was comparable if not higher that with Lipo 2000, and on a qualitative observation, the cells seemed healthier, we don’t estimate % death, but there were less cell floating/looking unhealthy in the LipoD293 transfection. We mixed 60 ul LipoD293 with 1ml DME no serum for 15 minutes at RT (mixed by bubbling), DNA with 1 ml of DME no serum was placed on a second tube for ~15 min as well (mixed by bubbling). We then transferred the DME/LipoD293 onto the 1ml of DME/DNA drop wise. We mixed by bubbling and incubated the now 2 ml for 15 min at RT. Mix was then added onto cells drop wise. The viral titers were equivalent to the lipo2K, 5-8x10*6
