At SignaGen Laboratories, lentivector cloning is performed using Gibson Assembly Cloning (GAC), traditional restriction enzyme (RE)–based cloning, or Golden Gate assembly to provide maximum flexibility, efficiency, and compatibility with diverse vector designs. These complementary cloning platforms allow seamless insertion of promoters, transgenes, selection markers, and regulatory elements into lentiviral transfer backbones while maintaining precise sequence architecture. Following cloning, all lentiviral plasmids undergo stringent verification by diagnostic restriction enzyme digestion to confirm correct insert size, orientation, and structural integrity.
Final construct validation is performed using whole-plasmid sequencing (WPS) on the Oxford Nanopore platform, enabling long-read, full-length coverage of promoters, coding regions, regulatory elements, and all junctions without gaps. This deep sequencing approach detects even subtle mutations, rearrangements, or recombination events that could compromise vector performance. Together, SignaGen’s integrated cloning and verification workflow ensures that every lentivector meets the highest standards of sequence accuracy, functional reliability, and reproducibility for high-titer, high-quality lentivirus production and downstream research applications.
