Custom synthesis of alternative adenoviral backbones is available when off-the-shelf human type C serotype 5 (Ad5) vectors do not fully meet your experimental requirements. While Ad5 remains the most widely used adenoviral backbone due to its well-characterized biology and robust packaging efficiency, certain applications may require alternative serotypes or engineered adenoviral genomes to achieve optimal performance.
It is important to note that the adenoviral genome is approximately 36 kb in length, making it significantly larger than most commonly synthesized plasmid constructs. The large genome size, combined with the presence of repetitive elements and sequence regions that can be unstable in standard cloning systems, presents substantial technical challenges for full adenoviral genome synthesis and assembly. Specialized design strategies and cloning approaches are therefore required to ensure genome stability and sequence integrity throughout the construction process.
To address these challenges, we offer custom construction of alternative adenoviral backbones tailored to your project. This may include the use of non-Ad5 serotypes, chimeric capsids, or engineered adenoviral genomes designed to improve tissue tropism, reduce pre-existing immunity, enhance transduction efficiency, or support specialized regulatory elements. Each backbone is synthesized, assembled, and sequence-verified to ensure high fidelity and reliability.
This capability enables researchers to access customized adenoviral systems better suited for specialized in vitro or in vivo applications when standard Ad5 vectors are not ideal.
