At SignaGen Laboratories, adenovirus purification and quality control are performed using rigorously optimized workflows to ensure consistency, potency, and suitability for downstream applications. Crude viral lysates are first clarified and purified by gel-filtration (size-exclusion chromatography) to efficiently remove cellular debris, proteins, and low-molecular-weight impurities, yielding in vitro–grade adenovirus with high recovery and reproducibility. For applications requiring higher purity, additional ultracentrifugation–based purification is applied to further enrich intact viral particles and reduce background contaminants.
Purified adenovirus preparations undergo comprehensive QC testing. Plaque-forming unit (PFU) titration is performed to quantify infectious viral particles, while transduction unit (TU) titration evaluates functional gene delivery efficiency in permissive cells. Additional QC assays may include physical particle titration, purity assessment, sterility testing, and identity confirmation. Together, these orthogonal purification and QC strategies ensure reliable adenovirus performance, accurate dosing, and reproducible results for gene overexpression, genome editing, and other research applications.
