Adenovirus Packaging & Multi-Round Amplification is conducted through a rigorously controlled workflow engineered to maximize viral yield, genetic integrity, and batch-to-batch reproducibility. The process begins with precise linearization of the adenoviral backbone plasmid to expose the inverted terminal repeats (ITRs), a critical prerequisite for efficient viral genome rescue. The linearized genome is then transfected into optimized Ad.MAX™ packaging cells that supply essential adenoviral functions in trans, enabling efficient initial virus assembly and release. Following primary rescue, the adenovirus undergoes carefully monitored, multi-round amplification to progressively increase viral titer while preserving genome stability and transgene fidelity. Each amplification cycle is performed under standardized, validated conditions to ensure consistent viral growth kinetics and minimize the risk of recombination or replication-competent adenovirus formation. This stepwise packaging and amplification strategy yields high-titer, high-quality adenoviral stocks suitable for robust gene overexpression, HDR donor delivery, and both in vitro and in vivo functional studies.
