At SignaGen Laboratories, Adenoviral Backbone Construction, Verification & QC is performed using a rigorous, Gateway®-based workflow to ensure accurate and reproducible adenovirus generation. Following successful Ad.MAX™ shuttle vector design, the transgene expression cassette is precisely transferred into the adenoviral backbone via Gateway® recombination, enabling high-efficiency, site-specific integration while preserving correct orientation and reading frame.
Each recombinant adenoviral backbone undergoes comprehensive verification using diagnostic restriction enzyme (RE) digestion to confirm structural integrity, insert size, and recombination accuracy. In parallel, whole-plasmid sequencing (WPS) is performed to provide base-level confirmation across the entire viral backbone, including the transgene cassette, recombination junctions, regulatory elements, and critical viral regions. This combined construction, verification, and quality control strategy minimizes production risk and ensures that only fully validated adenoviral backbones proceed to downstream adenovirus packaging, delivering reliable vector performance and consistent transgene expression.
