For AAV cis plasmids provided by the customer, SignaGen performs a rigorous incoming verification process to confirm plasmid integrity and ensure compatibility with downstream rAAV production. This process begins with SmaI restriction enzyme digestion, a critical quality control step specifically used to evaluate the presence, integrity, and correct configuration of both inverted terminal repeats (2× ITRs). Because intact ITRs are essential for AAV genome replication, packaging, and transduction efficiency, the resulting digestion pattern is carefully analyzed to detect deletions, rearrangements, or recombination events that may compromise vector performance. In parallel, Whole Plasmid Sequencing (WPS) is conducted to provide base-level confirmation of the entire cis plasmid, including both ITRs, the transgene cassette, promoters, regulatory elements, polyadenylation signals, and backbone regions. The combined use of SmaI digestion and WPS offers orthogonal validation of plasmid structure and sequence fidelity, significantly reducing production risk. Only cis plasmids that pass these verification criteria proceed to rAAV packaging, ensuring reliable vector yield, high titer, and consistent performance from customer-supplied constructs.
