Targeting testicular tissue with AAV is a specialized area of research focused on treating male infertility, testosterone deficiency (hypogonadism), and exploring the potential for germline editing. The testis is an “immune-privileged” site, but it is protected by the formidable Blood-Testis Barrier (BTB), which limits the access of systemic vectors to the developing sperm cells.
| Serotype | Primary Target Cells | Preferred Route | Key Characteristics & Notes |
| AAV8 | Leydig Cells, Sertoli Cells | IT | The Endocrine Specialist. • Highly efficient at transducing Leydig cells (which produce testosterone). • Often used in studies aiming to restore androgen levels. |
| AAV9 | Sertoli Cells, Leydig Cells, Myoid Cells | IT | The Best All-Rounder. • Exhibits the broadest tropism within the testis. • Excellent for targeting Sertoli cells, which act as the “nurses” for developing sperm. |
| AAV5 | Sertoli Cells | IT / Rete | Shows a strong preference for Sertoli cells over other cell types. • Frequently used in studies involving the secretion of factors into the seminiferous tubules. |
| AAV1 | Leydig Cells | IT | Similar to AAV8 but generally slightly lower in efficiency. Efficient for local protein production. |
| AAV2 | Variable/Low | IT | Generally considered less efficient than AAV8 or AAV9 for testicular tissue in vivo. |
Research into engineered capsids for the testis is less mature than for the CNS or Eye, but certain variants show promise for bypassing tissue barriers.
| Variant | Primary Target Cells | Preferred Route | Key Characteristics & Notes |
| AAV-DJ | Broad Testicular Cells | IT | High Yield. • A synthetic hybrid that shows extremely high transduction efficiency in testicular cell cultures and organoids. • Often used for ex vivo research. |
| AAV-Anc80 | Sertoli & Leydig Cells | IT | Ancestral Potency. • Like in other organs, this synthetic capsid shows robust expression and better stability within the complex testicular microenvironment. |
| AAV-PHP.B | Vasculature & Stroma | IV | While designed for the brain, its enhanced ability to cross endothelial barriers makes it slightly more effective than AAV9 for systemic access to the testis, though still limited by the BTB. |

The BTB is one of the tightest blood-tissue barriers in the body.
