Salivary glands are highly attractive targets for AAV-mediated gene therapy because they are encapsulated, easily accessible via the mouth, and can act as “bio-reactors” to secrete therapeutic proteins into both the saliva (for local oral/esophageal issues) and the bloodstream (for systemic diseases like diabetes or growth hormone deficiency).
| Serotype | Primary Target Cells | Preferred Route | Key Characteristics & Notes |
| AAV5 | Ductal & Acinar Cells | Retrograde | The Rodent Gold Standard. • Uses PDGFR (Platelet-Derived Growth Factor Receptor) for entry. • Shows significantly higher (5–10x) transduction efficiency than AAV2 in mice. • Note: Interestingly, efficiency is much lower in non-human primates (NHPs). |
| AAV2 | Ductal Cells (Striated/Intercalated) | Retrograde | The Clinical Benchmark. • The most extensively studied in humans (e.g., for Sjögren’s syndrome and radiation-induced xerostomia). • Uses HSPG (Heparan Sulfate Proteoglycan) as a receptor. • Provides stable, long-term expression in primates (>6 months). |
| AAV9 | Ductal Cells, Stroma | Retrograde | High Spread. • Highly efficient in rodents, similar to AAV5. • Known for its ability to escape the gland and enter the systemic circulation better than AAV2, which is useful if the goal is systemic protein delivery. |
| AAV4 | Ductal Cells (including Excretory) | Retrograde | Broad Ductal Tropism. • Transduces a wider variety of ductal segments (convoluted granular tubules) compared to AAV2/5. • Uses O-linked sialic acid for cell entry. |
| AAVRh10 | Acinar Cells | Retrograde | The Acinar Specialist. • One of the few serotypes that shows strong tropism for acinar cells (the cells that actually produce saliva) in rats and mice. • Superior to AAV2 in some NHP models. |
These are often selected for their ability to target the secretory acinar cells or to function better in human tissue than standard rodent-optimized serotypes.
| Variant | Primary Target Cells | Preferred Route | Key Characteristics & Notes |
| AAV44.9 | Acinar Cells (Parotid/Sublingual) | Retrograde | Novel Secretion Tool. • A newer isolate found to be highly effective at transducing acinar cells. • Results in robust secretion of the transgene product into both saliva and blood. |
| AAV12 | Salivary Epithelium | Retrograde | Antibody Evader. • Does not require sialic acid or heparin for entry. • Uniquely resistant to the neutralizing antibodies (NAbs) commonly found in human serum, making it a “cleaner” clinical candidate. |
| AAV-DJ | Broad Epithelium | Retrograde | High Output. • A synthetic hybrid that provides extremely high levels of protein production in salivary cells in vitro and in rodent models. |
If you are choosing a vector for a specific salivary application:
Most natural AAVs (especially AAV2) have a “Ductal Tropism.”
