Targeting the lymph nodes with AAV is a specialized field, often focused on vaccine development, cancer immunotherapy, or treating lysosomal storage diseases. Unlike other organs, the challenge here is not just reaching the tissue, but specifically transducing Antigen-Presenting Cells (APCs) like dendritic cells and macrophages.
| Serotype | Primary Target Cells | Preferred Route | Key Characteristics & Notes |
| AAV5 | Dendritic Cells (DCs), Macrophages | SC / IM | The Vaccine Specialist. • Exhibits higher natural tropism for human and mouse dendritic cells than most other serotypes. • Excellent for inducing robust T-cell and B-cell immune responses against a transgene. |
| AAV1 | Dendritic Cells, Myocytes | SC / IM | Robust Immune Induction. • Frequently used in genetic immunization (vaccines). • Highly efficient at transducing APCs in the skin and muscle that then migrate to the lymph nodes. |
| AAV9 | Stroma, Some Immune Cells | SC / IV | Broad & Diffuse. • Known for its ability to cross endothelial barriers. While it can reach lymph nodes, it is less “immune-cell specific” than AAV5 or AAV1. |
| AAV8 | Hepatocytes, Some APCs | SC | High Efficiency. • While primarily a liver vector, AAV8 has shown high transduction of the subcapsular sinus macrophages in lymph nodes following subcutaneous delivery. |
| AAV2 | APCs (Low efficiency) | Direct | The Reference. • Generally has lower transduction of immune cells compared to AAV1/5/6 unless delivered at extremely high titers. |
These are designed to enhance uptake by professional immune cells or shield the virus from neutralizing antibodies.
| Variant | Primary Target Cells | Preferred Route | Key Characteristics & Notes |
| AAV6.2 | Dendritic Cells, Airway Epithelium | SC / IN | A modified AAV6 that shows improved stability and higher transduction of dendritic cells, making it a strong candidate for immunotherapy. |
| AAV-DJ | Broad (including APCs) | SC / Direct | Synthetic Hybrid. • Outperforms most natural serotypes in in vitro DC transduction. • Useful for ex-vivo loading of dendritic cells before re-injecting them into the patient. |
| AAV-S663V | Antigen-Presenting Cells | IM / SC | The Proteasome Dodger. • A capsid mutation that prevents proteasomal degradation. • Increases the amount of intact virus reaching the lymph node, significantly boosting the immune response at lower doses. |

In most AAV therapies (like for Heart or Liver), the immune response is an enemy. In Lymph Node targeting, it is often the goal.
