| Promoter | Target Cell Type | Size (bp) | Origin | Key Characteristics |
| Ksp-cadherin | Renal Tubules | ~1.3 kb | Rabbit / Human | Kidney-specific cadherin. Highly restricted to the epithelial cells of the distal tubules and loops of Henle. |
| NPHS1 (Nephrin) | Podocytes | ~1.2 kb – 4.0 kb | Human / Mouse | Essential for the glomerular filtration barrier. Used to target the specialized cells that wrap around capillaries. |
| AQP2 | Collecting Duct | ~1.0 kb – 2.0 kb | Human / Mouse | Aquaporin 2. Specifically active in the principal cells of the collecting duct; regulated by vasopressin. |
1. Targeting the Filtration Barrier (NPHS1) Podocytes are terminally differentiated cells that are critical for preventing protein from leaking into the urine. The Nephrin (NPHS1) promoter is the primary tool for treating “podocytopathies.” Because podocytes do not divide, a single AAV or lentiviral delivery using a Nephrin promoter can provide very long-term therapeutic effects.
2. Tubular Specificity with Ksp-cadherin Ksp-cadherin (CDH16) is one of the most tissue-specific markers discovered for the kidney. Unlike many other “kidney” promoters that might show some leakiness in the liver or thyroid, Ksp-cadherin is strictly limited to the renal tubular epithelial cells. This makes it ideal for metabolic kidney disorders or polycystic kidney disease (PKD) research.
3. Water Balance and AQP2 The AQP2 promoter is unique because its activity is functionally linked to the body’s hydration status. It is restricted to the collecting ducts, which are the final site of urine concentration. Researchers use this promoter to study or treat Nephrogenic Diabetes Insipidus, where water reabsorption is impaired.
4. The “Size vs. Specificity” Trade-off Native kidney promoters are often quite large. For AAV applications, researchers frequently look for “minimal” enhancer/promoter combinations. For instance, a 1.2 kb fragment of the human NPHS1 promoter is often sufficient to drive podocyte-specific expression while leaving ~3.5 kb of space for the therapeutic gene.
