| Tissue/Cell Type | Promoter | AAV-Optimized Size | Target | Note on Longevity |
| Intestinal Epithelium | Villin | ~1.5 – 2.5 kb | All mature enterocytes | High turnover; expression is transient unless stem cells are hit. |
| Enterocytes | Fabp1 | ~1.0 – 1.5 kb | Small Intestine/Liver | Often includes a 3′ enhancer for potency. |
| Goblet Cells | MUC2 | ~1.5 – 2.0 kb | Mucus-secreting cells | Truncated versions used to target the colonic/intestinal barrier. |
| Stem Cells | Lgr5 | ~2.0 kb | Crypt Base Stem Cells | High value: Targeting these allows for permanent lineage tracing. |
| Stomach G-Cells | Gastrin | ~0.5 – 1.3 kb | Gastric Antrum | Smaller fragments are highly effective for endocrine targeting. |
Because enterocytes and goblet cells are shed so frequently, an AAV targeting mature cells (using Villin or Fabp1) will only show expression for about a week. To achieve long-term expression, you must target the Lgr5+ stem cells located at the base of the crypts.
If using Fabp1, be aware that it is also naturally expressed in the liver. To ensure your construct is truly GI-specific:
