Summary Table: Adipose-Specific Promoters & Sequence Sizes

PromoterBest AAV VersionOptimized SizeTargetAAV Serotype Note
mAdipoqTruncated Adipoq~2.1 kbMature WAT/BATMost validated for systemic AAV8 delivery.
hAdipoqHuman Adipo-e/p~1.1 kbMature WAT/BATUses a distal enhancer + proximal promoter. Very compact.
aP2 (Fabp4)mAP2.2 fragment~2.2 kbMature AdipocytesHigh expression but watch for liver/macrophage leakiness.
Ucp1mini-Ucp1~2.0 kbBAT/Beige FatEssential for thermogenic fat studies.
Plin1Proximal Plin1~1.0 kbMature AdipocytesVery small; ideal if your transgene (GOI) is large (>2 kb).
SyntheticPpar$\gamma$-RE x3< 0.5 kbMature AdipocytesUltra-short; uses tandem repeats of binding elements.

Critical Considerations for AAV Production

1. Managing “Leakiness” with miR-122

Even with adipose-specific promoters, AAVs (especially AAV8/9) have a high natural tropism for the liver. To ensure your gene is only in fat:

  • Add miR-122 target sites: Inserting 3–4 copies of the miR-122 binding site in the 3′ UTR of your vector will cause the liver to degrade your transgene’s mRNA, effectively “shaping” the expression to be fat-exclusive.

For adipose tissue, the choice of capsid (serotype) is as important as the promoter:

  • AAV8: Generally considered the gold standard for systemic (IV) delivery to both White Adipose Tissue (WAT) and Brown Adipose Tissue (BAT).
  • AAV9: Also highly effective for adipose, though it has higher cardiac and CNS cross-reactivity.
  • AAV-Rec2: A recently engineered synthetic serotype showing high efficiency specifically in adipocytes.
