PepJet™ In Vivo DNA Transfection Reagent is a next-generation cationic peptide–based delivery system engineered for highly efficient plasmid DNA transfection in vivo. The core active component is a synthetic 28-amino-acid peptide originally identified through screening as a potent nucleic acid delivery vehicle and subsequently optimized using sequence alignment, molecular modeling, and computational analyses inspired by naturally occurring viral protein architectures. The peptide was chemically synthesized, structurally refined, and extensively validated through iterative in vitro and in vivo studies to maximize gene transfer efficiency.

To improve systemic stability and biological performance, a proprietary shielding peptide is covalently incorporated into the delivery platform. This modification reduces nonspecific interactions with serum proteins and other biological components following systemic administration, thereby enhancing the formation, stability, and biodistribution of PepJet™/DNA complexes. In animal studies, PepJet™ has demonstrated efficient plasmid DNA delivery and robust transgene expression in multiple organs, including the lung, liver, spleen, and kidney, following systemic administration.

Storage Conditions
Store at 4°C. When maintained under the recommended storage conditions, PepJet™ DNA In Vivo Transfection Reagent is stable for at least 24 months.

Performance Evaluation
The in vivo transfection efficiency of PepJet™ reagent was evaluated in mice using a plasmid encoding Renilla luciferase as a reporter gene. Following systemic administration of PepJet™/DNA complexes, transgene expression levels were quantified in various organs to assess tissue distribution and gene delivery efficiency. PepJet™ mediated robust Renilla luciferase expression in the lung, liver, spleen, and kidney, demonstrating its effectiveness as a systemic plasmid DNA delivery platform.
