GenJet™ DNA In Vitro Transfection Reagent for U2OS Cells is pre-optimized for efficient DNA transfection of U2OS cells. U2OS is a human osteosarcoma cell line originally established from the tibial tumor tissue of a 15-year-old female patient and has become one of the most widely used cell models in cancer biology and molecular genetics. U2OS cells exhibit an adherent epithelial-like morphology and are extensively utilized in studies of cell cycle regulation, DNA damage response, genome stability, signal transduction, gene expression, and cancer therapeutics. The cell line retains wild-type p53 function, making it particularly valuable for investigating mechanisms of tumor suppression and cellular stress responses.

The transfection protocol described below has been optimized to achieve maximal transfection efficiency in U2OS cells while maintaining excellent cell viability. One milliliter of GenJet™ reagent is sufficient for approximately 300–600 transfections in 24-well plates or 150–300 transfections in 6-well plates.

Storage Conditions
Store at 4°C. When maintained under recommended storage conditions, the reagent remains stable for at least 24 months.

U2OS cells were cultured according to ATCC-recommended growth conditions and transfected with the GFP expression plasmid pEGFP-N3 using GenJet™ reagent.
