Description:
Ad-CMV-Mito-PA-GFP is a recombinant human adenovirus type 5 (ΔE1/ΔE3) that expresses a mitochondrial-targeted photoactivatable green fluorescent protein (Mito-PA-GFP) under the control of the CMV promoter. The mitochondrial targeting sequence (MTS) derived from subunit VIII of human cytochrome c oxidase is fused to the N-terminus of PA-GFP, directing the fusion protein specifically to mitochondria in transduced cells. Upon photoactivation with 405-nm light, PA-GFP undergoes an approximately 100-fold increase in green fluorescence when subsequently excited at 488 nm, with the activated signal remaining stable for several days under aerobic conditions. This vector enables selective photoactivation and long-term tracking of individual mitochondria, making it a powerful tool for studying mitochondrial dynamics, trafficking, fusion/fission, and turnover in living cells. Supplied in a ready-to-use format for in vitro and in vivo applications.
