Ad-CMV-SaCas9-CMV-GFP is a pre-packaged, replication-deficient human adenovirus type 5 (E1/E3 deleted) designed for efficient CRISPR-mediated genome editing and real-time monitoring of transduction efficiency. This vector co-expresses a codon-optimized Staphylococcus aureus Cas9 (SaCas9) nuclease and GFP reporter, each driven by independent CMV promoters to ensure robust expression across a wide range of mammalian cell types.
SaCas9 is a compact CRISPR nuclease that recognizes a longer PAM sequence (NNGRRT), offering enhanced targeting specificity and making it particularly suitable for applications with vector size constraints. The co-expressed GFP enables convenient visualization and enrichment of successfully transduced cells.
This adenoviral vector provides high transduction efficiency in both dividing and non-dividing cells, supporting transient genome editing, functional genomics studies, and in vivo applications. Guide RNAs (gRNAs) can be delivered separately or via compatible vectors to achieve precise, sequence-specific genome modification.
