Ad-CMV-Cas9(D10A)-T2A-DasherGFP

Product Category: Human Adenovirus Type 5 (ΔE1/E3) | Ad-CMV-Cas9(D10A)-T2A-GFP | Ad-CMV-Cas9(D10A)-T2A-DasherGFP
Description:
Ad-CMV-Cas9(D10A)-T2A-DasherGFP is a recombinant, E1/E3-deleted human adenovirus serotype 5 vector engineered to express a codon-optimized Streptococcus pyogenes Cas9(D10A) nickase under the control of the CMV immediate early promoter. The Cas9(D10A) variant contains a point mutation in the RuvC nuclease domain, converting the wild-type Cas9 into a DNA nickase that introduces single-strand breaks (nicks) instead of double-strand breaks.
This vector co-expresses DasherGFP via a self-cleaving T2A peptide, enabling real-time monitoring of transduction efficiency and transgene expression in mammalian cells. The Cas9 nickase can be used in combination with paired guide RNAs (gRNAs) to generate offset single-strand nicks on opposite DNA strands, resulting in high-specificity genome editing with significantly reduced off-target effects compared to wild-type Cas9.
The adenoviral delivery system enables efficient transduction across a broad range of dividing and non-dividing cell types, providing robust transient expression without genomic integration. This makes the vector particularly suitable for applications requiring precise genome modification, reduced off-target activity, or short-term Cas9 expression.
Key Features:
