Description:
Ad-CMV-3xFLAG-SpCas9-P2A-PuroR is a pre-packaged, replication-deficient human adenovirus type 5 (E1/E3 deleted) designed for efficient CRISPR-based genome editing in mammalian cells. This vector expresses a codon-optimized Streptococcus pyogenes Cas9 (SpCas9) fused with an N-terminal 3×FLAG tag for convenient detection and validation.
The Cas9 nuclease is co-expressed with a puromycin resistance gene (PuroR) via a self-cleaving P2A peptide, enabling antibiotic selection of successfully transduced cells. Expression is driven by the strong CMV promoter to ensure robust Cas9 production across a wide range of cell types.
This adenoviral vector provides high transduction efficiency in both dividing and non-dividing cells, making it suitable for transient genome editing applications, functional genomics studies, and CRISPR screening workflows. Guide RNAs (gRNAs) can be delivered separately or via compatible vectors to achieve targeted gene modification.
Supplied in a ready-to-use format for in vitro and in vivo research applications.
