AAV9-CBh-TAU(P301L-S320F)-FLAG is a pre-made recombinant adeno-associated virus (rAAV) packaged in serotype 9 (AAV9 capsid with AAV2 ITRs), designed for expression of a mutant human tau protein harboring the P301L and S320F substitutions. Expression is driven by the hybrid CBh promoter, enabling strong and stable transgene expression across a broad range of mammalian cell types, including neurons in the central nervous system.

The encoded tau protein contains two disease-associated mutations: P301L, a well-characterized mutation linked to frontotemporal dementia, and S320F, which has been reported to enhance tau aggregation and accelerate pathology. Together, these mutations provide a robust model for studying tau misfolding, aggregation, and neurodegenerative processes.

A C-terminal FLAG tag is fused to the tau protein to facilitate detection, purification, and downstream biochemical analyses using anti-FLAG antibodies.

This vector is well-suited for in vivo and in vitro applications investigating tauopathies, including mechanisms of aggregation, propagation, and therapeutic intervention strategies.
