Description:
AAV(DJ)-Syn-DIO-NaChBac-dTomato is a pre-made recombinant adeno-associated virus (rAAV) packaged in AAV-DJ capsid and designed for Cre-dependent expression of NaChBac and the fluorescent reporter dTomato in neurons. The Syn (synapsin) promoter drives preferential neuronal expression, while the DIO (Double-floxed Inverted Open reading frame) configuration restricts functional transgene expression to Cre recombinase-positive cells.

AAV-DJ is a synthetic chimeric AAV capsid generated through DNA shuffling of multiple naturally occurring AAV serotypes. It exhibits broad tropism and efficient transduction of a variety of mammalian cell types, making this vector suitable for a broad range of neuroscience and gene delivery applications.

NaChBac is a bacterial voltage-gated sodium channel that is commonly used as a genetic tool to enhance neuronal membrane excitability. In Cre-expressing cells, Cre-mediated recombination switches the DIO cassette into the transcriptionally active orientation, enabling expression of NaChBac together with dTomato. NaChBac expression can enhance neuronal excitability and firing, while dTomato provides a red fluorescent marker for visualization and identification of transduced, Cre-positive neurons.

AAV(DJ)-Syn-DIO-NaChBac-dTomato is particularly suitable for Cre-dependent manipulation of neuronal excitability, functional analysis of genetically defined neuronal populations, neural circuit studies, investigation of the physiological and behavioral consequences of neuronal activation, and fluorescent identification of targeted neurons. It can be used with Cre-driver animal models or in combination with Cre-expressing viral vectors to achieve cell type- and circuit-specific manipulation of neuronal activity.
