AAV-S5E2-hM4D(Gi)-P2A-dTomato (Serotype 1)

Product Category: Pre-made recombinant AAV; Chemogenetic AAV; DREADD AAV; PV Interneuron-Targeting AAV; AAV1-S5E2-hM4D(Gi)-P2A-dTomato
Description:
AAV1-S5E2-hM4D(Gi)-P2A-dTomato is a pre-packaged recombinant adeno-associated viral (rAAV) vector in serotype 1 designed for cell type-selective chemogenetic inhibition of parvalbumin-positive (PV+) interneurons.
The vector utilizes the S5E2 enhancer, a compact regulatory element derived from the SCN1A/Scn1a locus that has been developed for preferential transgene expression in PV+ cortical interneurons. The cell type selectivity of S5E2 makes this vector particularly useful for targeted manipulation of PV+ inhibitory neural circuits.
S5E2 drives expression of hM4D(Gi), an inhibitory Gi-coupled DREADD receptor. Upon activation with an appropriate DREADD agonist, hM4D(Gi) activates Gi-mediated signaling to suppress neuronal excitability, enabling reversible chemogenetic inhibition of S5E2-targeted PV+ interneurons.
A P2A self-cleaving peptide enables co-expression of hM4D(Gi) and the fluorescent reporter dTomato from the same transcript. dTomato provides convenient visualization and identification of transduced cells.
The expression cassette also incorporates a chimeric intron, WPRE, and bGH poly(A) signal to support efficient transgene expression and RNA processing. The cassette is flanked by AAV2 ITRs and packaged in the AAV1 capsid.
