Product Category: Ready to package recombinant AAV; Activity-dependent AAV; RAM-d2TTA/TRE AAV; Neuronal Activity Reporter AAV; AAV-RAM-d2TTA::TRE-NLS-tdTomato-WPRE
Description:
AAV-RAM-d2TTA::TRE-NLS-tdTomato-WPRE is a pre-packaged recombinant adeno-associated virus (rAAV) designed for activity-dependent labeling of activated neuronal populations. The vector utilizes the RAM (Robust Activity Marking) regulatory system, in which neuronal activity drives expression of the destabilized tetracycline-controlled transactivator d2TTA. d2TTA subsequently activates the TRE (tetracycline response element) to drive expression of nuclear-localized tdTomato (NLS-tdTomato).
This activity-dependent transcriptional architecture enables fluorescent labeling of neurons that are active during a defined experimental period. Nuclear localization of tdTomato facilitates visualization, identification, and quantification of activity-tagged cells, while WPRE enhances transgene expression.
Because the RAM-d2TTA/TRE system is tetracycline-responsive, the temporal window of activity-dependent labeling can be experimentally controlled using tetracycline-class compounds such as doxycycline, depending on the specific experimental design.
