Product Category: Pre-made recombinant AAV; Activity-dependent AAV; RAM-d2TTA/TRE AAV; Neuronal Activity Reporter AAV; AAV(PHP.eB)-RAM-d2TTA::TRE-NLS-dTomato-WPRE
Description:
AAV(PHP.eB)-RAM-d2TTA::TRE-NLS-dTomato-WPRE is a pre-packaged recombinant adeno-associated virus (rAAV) designed for activity-dependent labeling of activated neuronal populations. The vector utilizes the RAM (Robust Activity Marking) regulatory system, in which neuronal activity drives expression of the destabilized tetracycline-controlled transactivator d2TTA. d2TTA subsequently activates the TRE (tetracycline response element) to drive expression of nuclear-localized dTomato (NLS-dTomato).
This activity-dependent transcriptional architecture enables fluorescent labeling of neurons that are active during a defined experimental period. Nuclear localization of dTomato facilitates visualization, identification, and quantification of activity-tagged cells, while WPRE enhances transgene expression.
The vector is packaged in the PHP.eB capsid, an engineered AAV capsid developed for efficient CNS transduction in susceptible mouse strains. This vector is particularly useful for neuronal activity mapping, activity-dependent circuit labeling, identification of neuronal ensembles, behavioral neuroscience, and studies of experience-dependent neural activation.
Because the RAM-d2TTA/TRE system is tetracycline-responsive, the temporal window of activity-dependent labeling can be experimentally controlled using tetracycline-class compounds such as doxycycline, depending on the specific experimental design.
