Description:
AAV(BI30)-ICAM2-EGFP-3×miR122 is a pre-made recombinant adeno-associated virus engineered to express enhanced green fluorescent protein (eGFP) under the control of the mouse ICAM2 promoter, enabling preferential transgene expression in vascular endothelial cells. Three tandem miR-122 target sequences (3×miR122) are incorporated into the 3′ untranslated region to suppress transgene expression in hepatocytes, thereby reducing off-target liver expression following systemic administration. Packaged in the AAV(BI30) capsid, this vector exhibits enhanced tropism for central nervous system (CNS) endothelial cells in permissive mouse strains, making it an excellent tool for studies of the blood-brain barrier (BBB), cerebrovascular biology, endothelial gene function, and CNS-targeted gene delivery.

This vector is produced using our helper-free AAV packaging system, purified by CsCl gradient ultracentrifugation, and supplied in a ready-to-use format at >1 × 10¹³ VG/mL (in vivo grade). Each lot undergoes rigorous quality control, including vector genome titration by qPCR using an ATCC reference AAV standard, to ensure high purity, consistency, and reliable performance. The ICAM2 promoter provides endothelial-enriched transgene expression, while the 3×miR122 cassette further minimizes hepatic expression through endogenous miR-122-mediated post-transcriptional silencing. The eGFP reporter enables convenient visualization and quantification of transduced cells by fluorescence microscopy, flow cytometry, or histological analysis, making this vector particularly well suited for selective labeling of CNS endothelial cells after systemic delivery.
