Description:
AAV9-hSyn-TAU(P301L) is a pre-made recombinant adeno-associated virus packaged in serotype 9 (AAV9 capsid with AAV2 ITRs) designed for neuron-specific expression of the human tau protein harboring the P301L mutation. The P301L mutation in the MAPT gene is a well-characterized pathogenic variant associated with tauopathies, including frontotemporal dementia with parkinsonism linked to chromosome 17 (FTDP-17), and is widely used to model neurodegenerative processes.

Expression is driven by the human synapsin (hSyn) promoter, enabling selective and robust transgene expression in neuronal populations. Following transduction, the mutant tau protein undergoes abnormal phosphorylation and aggregation, leading to neurofibrillary tangle formation, synaptic dysfunction, and progressive neuronal degeneration.

AAV9-hSyn-TAU(P301L) is an ideal tool for in vivo modeling of tau pathology, allowing researchers to investigate disease mechanisms, neuronal toxicity, and the spread of tau aggregates across neural circuits.
