Description:
AAV1-hSyn-RiboL1 is a pre-packaged recombinant adeno-associated viral (rAAV) vector in serotype 1 designed as a control vector for AAV1-hSyn-RiboL1-jGCaMP8s. It contains the same hSyn promoter and RiboL1 element but lacks the jGCaMP8s calcium indicator, making it an ideal matched control for experiments using AAV1-hSyn-RiboL1-jGCaMP8s.
The human synapsin (hSyn) promoter provides neuron-specific expression, while RiboL1 serves as a soma-targeting element for preferential localization to neuronal cell bodies. The vector also contains WPRE and a bGH poly(A) signal, with the expression cassette flanked by AAV2 ITRs and packaged in the AAV1 capsid.
By providing a matched AAV1 vector without jGCaMP8s, AAV1-hSyn-RiboL1 serves as an excellent negative control for distinguishing jGCaMP8s-dependent calcium signals and biological effects from those associated with AAV transduction or other vector components.
