AAV9-hSyn-GRAB-DA2m is a pre-made recombinant adeno-associated virus (rAAV) packaged in serotype 9 capsid and designed to express the second-generation, medium-affinity genetically encoded dopamine (DA) sensor GRAB-DA2m in neurons. The human synapsin (hSyn) promoter drives preferential neuronal expression, making this vector well suited for monitoring dopamine dynamics in neuronal populations.

GRAB-DA2m is a genetically encoded fluorescent dopamine sensor engineered to report changes in extracellular dopamine through dopamine-dependent changes in fluorescence. The medium-affinity DA2m variant provides sensitive detection of physiologically relevant dopamine fluctuations while maintaining the temporal resolution needed to monitor dynamic dopaminergic signaling.

AAV9 provides efficient neuronal transduction in many regions of the central and peripheral nervous systems and is widely used for in vivo neuroscience applications. Following transduction, GRAB-DA2m is expressed on the neuronal plasma membrane, enabling real-time optical monitoring of extracellular dopamine release and dynamics in cultured neurons, brain slices, and living animals.
