AAV9-CBh-TAU(P301L-S320F)-mCherry is a pre-made recombinant adeno-associated virus (rAAV) packaged in serotype 9 (AAV9 capsid with AAV2 ITRs), designed for expression of a mutant human tau protein harboring the P301L and S320F substitutions, along with a fluorescent reporter. Expression is driven by the hybrid CBh promoter, enabling strong and stable transgene expression across a broad range of mammalian cell types, including neurons in the central nervous system.

The TAU(P301L-S320F) variant combines two disease-associated mutations: P301L, a well-characterized mutation linked to frontotemporal dementia, and S320F, which has been shown to enhance tau aggregation and accelerate pathological progression. This dual-mutant construct provides a robust model for studying tau misfolding, aggregation, and neurodegeneration.

The inclusion of mCherry allows direct visualization of transduced cells and facilitates tracking of expression and distribution in vitro and in vivo.

This vector is well-suited for studies of tauopathies, including mechanisms of aggregation, propagation, and evaluation of therapeutic strategies.
