Description:
AAV(BI30)-CAG-tdTomato-DIO-shRNAmir(SCRM) is a pre-made recombinant AAV packaged in the BI30 capsid designed to provide constitutive fluorescent labeling alongside Cre-dependent expression of a control shRNAmir cassette. The CAG promoter drives tdTomato continuously, enabling robust visualization and tracking of transduced cells regardless of Cre status. In the same vector, a double-floxed inverted orientation (DIO) element controls expression of a scrambled (SCRM) shRNAmir embedded in a miR-30 backbone, so the shRNAmir is activated only after Cre-mediated recombination. The miR-30–based shRNAmir architecture leverages endogenous microRNA processing pathways (Drosha/Dicer), supporting efficient, stable, and typically low-toxicity expression of the hairpin compared with some traditional shRNA formats. Because the shRNAmir sequence is scrambled and non-targeting, this construct serves as a Cre-conditional negative control for RNAi experiments while maintaining always-on tdTomato as a transduction reporter.
