AAV(PHP.eB)-CAG-GFP-httQ72 is a pre-made recombinant adeno-associated virus (rAAV) packaged with the engineered PHP.eB capsid (with AAV2 ITRs) designed for robust expression of a GFP-tagged huntingtin (HTT) protein fragment containing 72 CAG repeats (encoding glutamine, Q), representing a pathogenic polyglutamine (polyQ72) expansion associated with Huntington’s disease. Expression is driven by the strong, ubiquitous CAG promoter (CMV enhancer/chicken β-actin promoter), enabling high-level transgene expression across a wide range of mammalian cell types, with particularly efficient transduction in the central nervous system (CNS).

The PHP.eB capsid enables enhanced blood–brain barrier (BBB) penetration and widespread neuronal transduction following systemic delivery in mice. The GFP fusion allows real-time visualization of mutant HTT protein localization, aggregation, and inclusion body formation in both in vitro and in vivo systems. The expanded polyQ72 tract promotes misfolding and aggregation, recapitulating key pathological features of Huntington’s disease.
