Description:
AAV9-CAG-Flex-GFP-Tau(P301L) is a pre-made recombinant adeno-associated virus (rAAV) packaged in serotype 9 (AAV9 capsid with AAV2 ITRs), designed for Cre-dependent expression of a GFP-tagged human tau protein carrying the pathogenic P301L mutation. Expression is driven by the strong and ubiquitous CAG promoter, enabling robust transgene expression following Cre-mediated recombination.

This vector utilizes a FLEX (DIO, double-floxed inverted open reading frame) design, in which the GFP-Tau(P301L) cassette is inverted and flanked by heterotypic loxP/lox2272 sites. In the presence of Cre recombinase, the transgene is flipped into the correct orientation and permanently activated, allowing precise spatial and cell type-specific control of expression.

The P301L mutation in tau is a well-characterized pathogenic variant associated with frontotemporal dementia and is widely used to model tau aggregation, neurofibrillary tangle formation, and neurodegeneration. The GFP fusion enables direct visualization of tau expression, localization, and aggregation dynamics in live cells and tissues.

This vector is well-suited for in vivo and in vitro studies of tau pathology with cell type-specific targeting, particularly in transgenic Cre-expressing models.
