Conventional cationic transfection reagents provide high gene delivery efficiency in vitro but are poorly suited for systemic in vivo applications because they are rapidly cleared from circulation and tend to accumulate in the lungs through nonspecific interactions with blood components.
To address this limitation, we developed a modified GenJet™ Plus reagent by covalently attaching a shielding peptide (SP) to its neutral lipid backbone. This shielded formulation prevents erythrocyte aggregation, prolongs circulation after intravenous administration, and significantly enhances transgene expression in the lung, spleen, and liver compared with the original GenJet™ Plus reagent.
