SignaGen offers a comprehensive portfolio of pre-manufactured viral vectors designed for tightly regulated, inducible gene expression in mammalian systems. In parallel, we provide full-service custom vector engineering and production using adenoviral, adeno-associated viral (AAV), and lentiviral platforms to generate inducible expression constructs optimized for specific promoters, regulatory elements, and transgenes of interest. Our scientific support team delivers detailed vector maps, induction kinetics, and empirically guided dosing recommendations to suppress basal promoter activity and minimize off-target or background expression (“leakiness”). These measures enable high dynamic range, reproducible induction, and ensure experimental fidelity in studies requiring precise temporal and quantitative control of gene expression.
The Tet-On/Off system uses doxycycline to reversibly control gene expression, enabling precise induction or repression of a transgene in a dose-dependent manner. The Cumate–CymR system provides an orthogonal, small-molecule–regulated switch based on bacterial repressor biology, offering tight control and compatibility with Tet systems for dual or multiplexed regulation
Cre and Flpo are site-specific recombinase systems that act as precise “genetic scissors” to control gene expression with high spatial and temporal resolution. Cre recombinase recognizes loxP sites, while Flpo—an optimized variant of the yeast-derived Flp—targets FRT sites; upon expression, these enzymes mediate deletion, inversion, or translocation of DNA segments flanked by their respective recognition sites. Using both systems together, an approach known as intersectional genetics, allows researchers to manipulate only those cells defined by the overlap of two markers (for example, cells expressing both Gene A and Gene B). This dual-recombinase strategy is widely used for generating conditional knockout models, mapping and labeling neuronal circuits, and dissecting complex developmental lineages with minimal off-target effects on the organism
