Bioluminescence is widely used for in vivo imaging because it provides an exceptionally high signal-to-noise ratio. Mammalian tissues do not produce endogenous bioluminescence, resulting in essentially zero background signal
Fluorescent imaging requires an external excitation light source (such as a laser or LED) to stimulate the reporter protein. A major limitation for in vivo applications is tissue autofluorescence, in which endogenous biomolecules emit background signal—particularly in the green spectrum—thereby reducing sensitivity
Genetically encoded calcium indicators (GECI) like GCaMP6 & GCaMP8, etc, produce fluorescence in response to intracellular Ca²⁺ binding. Unlike synthetic dyes, GCaMP can be stably expressed from viral vectors or transgenes, enabling long-term, cell-type–specific monitoring of calcium dynamics. In neuroscience, GCaMP is widely used to visualize neuronal firing and synaptic activity in real time in living animals.
