AAV8 vs. AAV(Rec2): Which Capsid Is Better for Adipose Tissue Gene Delivery?

Selecting the right AAV serotype is critical for achieving efficient gene transfer to adipose tissue. While AAV8 has been widely used for metabolic research, AAV(Rec2) has emerged as an engineered capsid with significantly improved adipose tropism.

Key Comparison

🧬 AAV8

  • Well-established serotype for in vivo studies
  • Efficient transduction of both white and brown adipose tissue
  • High liver transduction following systemic administration
  • Readily available for many research applications

🧬 AAV(Rec2)

  • Engineered specifically for adipose tissue targeting
  • Superior transduction efficiency in both white adipose tissue (WAT) and brown adipose tissue (BAT)
  • Higher adipose specificity than conventional AAV8
  • Successfully validated for adipose-specific functional gene delivery in mouse models

Which Capsid Should You Choose?

✅ Local adipose depot injection (WAT or BAT): AAV(Rec2) is generally the preferred choice because it consistently provides higher transduction efficiency and stronger transgene expression than AAV8.

✅ Intraperitoneal (i.p.) administration: AAV(Rec2) has demonstrated efficient targeting of visceral adipose tissue while reducing off-target expression compared with conventional serotypes.

✅ Systemic intravenous (IV) administration: Both AAV8 and AAV(Rec2) can transduce adipose tissue, but significant liver transduction may still occur. Tissue-specific promoters or liver de-targeting strategies (e.g., miR-122 target sequences) are recommended when adipose-specific expression is desired.

Bioluminescence Imaging

For noninvasive monitoring of adipose transduction, luciferase reporters provide a convenient solution.

  • Firefly luciferase (fLuc): Reliable and widely used.
  • AkaLuc: Offers substantially higher sensitivity and brighter signals, particularly for deep-tissue imaging, making it an excellent choice for adipose imaging studies.

At SignaGen Laboratories, we offer both pre-made AAV8 luciferase vectors and custom-packaged AAV(Rec2) vectors to support adipose tissue research.
