Targeting the pancreas is often divided into two distinct goals: reaching the endocrine cells (the Islets of Langerhans, like alpha and beta cells for diabetes) or the exocrine cells (acinar cells for pancreatitis or cystic fibrosis).
For the pancreas, the route of administration is the primary driver of success, as systemic IV delivery often sees most of the virus sequestered by the liver.
| Serotype | Primary Target | Tropism Level | Optimal Route | Notes |
| AAV8 | Beta cells & Acinar | High | Intraperitoneal / IV | The most widely used natural serotype for the pancreas; targets both endocrine and exocrine cells. |
| AAV9 | Acinar Cells | Moderate/High | Systemic (IV) | Excellent for global distribution; generally outperforms AAV8 in total acinar cell transduction via the bloodstream. |
| AAV6 | Beta cells | Very High | Retrograde Ductal | When injected directly into the pancreatic duct, AAV6 shows superior efficiency in targeting islets/beta cells. |
| AAV2 | Islets & Acinar | Moderate | Local / Direct | The “standard” vector; effective for direct pancreatic injection but less efficient than AAV8 or AAV9 systemically. |
| AAV4 | Beta cells | Moderate | Systemic (IV) | Recently identified in “atlas” studies as having a unique preference for pancreatic beta cells in certain mouse models. |
| AAV-KP1 | Human Islets | Ultra High | Local / Ex vivo | Synthetic; engineered for human kidney/pancreas. Outperforms AAV8 by ~10x in human islet cells. |
| AAV-DJ | Pancreatic Tissue | High | Local Injection | A synthetic chimera often used in research for its high potency, though limited by high liver “leakage” if given systemically. |
Because the pancreas is highly vascularized, the delivery method determines which cells the AAV actually “sees”:
To ensure the gene only functions in the correct pancreatic cell type (vital for diabetes research):
If your research involves human islet transplantation or Type 1 Diabetes, AAV-KP1 is currently the state-of-the-art synthetic variant. It was developed through directed evolution to specifically solve the problem of low transduction in human pancreatic endocrine cells.
